چکیده

A13STRACT.--Genomic DNA libraries of the Barn Swallow (Hirundo rustica) and the Pied Flycatcher (Ficedula hypoleuca) were screened for the presence of dinucleotide microsatellite repeats. Two thymine-guanine repeats, "(TG),," and two thymine-cytosine repeats, "(TC),," were isolated and sequenced from the two species, respectively. Polymerase-chain-reaction (PCR) analysis of 25 unrelated Barn Swallows and 10 unrelated Pied Flycatchers revealed 3 to 15 alleles per locus and heterozygosities in the range of 0.46 to 0.89. Mendelian inheritance was confirmed for all four loci in 10 Pied Flycatcher and 2 Barn Swallow families comprising a total of 240 meioses. The occurrence of nonparental alleles in offspring from two Barn Swallow families was consistent with extrapair fertilization as revealed by a parallel DNAfingerprinting analysis. A DNA amount approximately corresponding to 0.01/•1 blood was used for PCR analysis. DNA was also prepared from feathers and approximately 1% of a preparation from a single remex or rectrix was sufficient for PCR amplification. One of the primer pairs from the Barn Swallow also amplified a polymorphic locus in the House Martin (Delichon urbica) and in the Bank Swallow (R. riparia). Received 3 October 1991, accepted 16 June 1992.

Isolation and Characterization of Microsatellite Markers from Endangered Species (Camelus bactrianus)

Iranian bactrian camel (Camelus bactrianus) is an endangered livestock breed with distribution in northwest of Iran. Microsatellites are a powerful marker for animal genetic and cell line identification and population genetic study. In this study, after producing more than 40 Camelus bactrianus fibroblast cell lines, microsatellites loci from the genome of Iranian Camelus bactrianus cell lines ...

Genetic Heterogeneity among Leishmania major Isolates in Iran Determined by Restriction Fragment Length Polymorphism (RFLP) and Multilocus Microsatellite Typing (MLMT)

Background & Aims: In recent years, molecular methods for characterizing genetic heterogeneity have found a major place in modern approaches. In this study, two different molecular techniques including Restriction Fragment Length Polymorphism (RFLP) and Multi Locus microsatellite typing (MLMT) were carried out in order to evaluate genetic heterogeneity among isolates of Leishmania major in Iran...

Polymerase chain reaction for the detection and differentiation of Marek’s disease virus strains MDV-1 and HVT

Marek’s disease (MD) is a lymphoproliferative disease of chickens characterized by lymphocyticinfiltration of various organs. The present study was an attempt to use polymerase chain reaction (PCR) tooptimize a rapid and reliable assay for detection of MDV genome. Detection of serotype 1 of MDV (MDV-1)was confirmed by presence of a 200 bp DNA fragment as a PCR product. Differentiation of MDV-1 ...

DETECTION AND RESTRICTION ANALYSIS OF C YTOMEGALOVIRUS DNA PERSISTING IN HUMAN ATHEROSCLEROTIC PLAQUES USING POLYMERASE CHAIN REACTION

The polymerase chain reaction (PCR) as applied to detection of a foreign DNA in clinical specimens could provide a sensitive instrument for rapid detection of viral DNA persisting in tissues of patients suspected of latent infection. Human cytomegalovirus (HCMV) DNA was found in arterial plaques of patients with atherosclerotic lesions using a PCR assay with nested primer oligonucleotides ...

Molecular detection and characterization of beak and feather disease virus in psittacine birds in Tehran, Iran

Beak and feather disease virus (BFDV), a member of genus circovirus, is a small, non-enveloped, single stranded DNA virus. Although BFDVs are among the most well studied circoviruses, there is little to no information about BFDVs in Iran. The aim of the present study was to detect and identify BFDV molecules from the birds referred to the avian clinic of The Faculty of Veterinary Medicine, Tehr...
